Recombinant GH3 β-glucosidase stimulated by xylose and tolerant to furfural and 5-hydroxymethylfurfural obtained from Aspergillus nidulans

In this work, authors cloned and overexpressed the β-glucosidase gene from Aspergillus nidulans FGSC A4 in A. nidulans A773. The recombinant enzyme (schematic representation its 3D structure illustrated above) retained 100% bioactivity after 24 h of incubation at 50 ºC and pH 6.0. The application of this enzyme product after purification was demonstrated in the saccharification of the tropical grass Panicum maximum, which led to a 48% increase in glucose release after 24 h. Thus, this recombinant enzyme is promising as part of a multi-enzyme cocktail for the efficient hydrolysis of lignocellulosic biomass in a biorefinery.
