miR-10b regulates EMT in human NPC cells. A. Cell morphology of HNE1 cells following transfection with miR-10b mimics and in HNE1/DDP cells following transfection with miR-10b inhibitors (images acquired at 200× magnification). B. Western blot analysis demonstrating that miR-10b mimics reduce E-cadherin protein abundance but enhanced vimentin and MMP9 protein abundance in HNE1 cells. In contrast, miR-10b inhibitors promoted E-cadherin protein abundance but decreased vimentin and MMP9 protein abundance in HNE1/DDP cells. C. qRT-PCR analysis demonstrating that expression of E-cadherin was down-regulated, whereas vimentin and MMP9 expression were up-regulated in HNE1 cells following transfection of miR-10b mimics. The relative amount of mRNA was normalized to GAPDH (mean ± SEM, *P<0.05 compared to NC).
