Discovery of a Simple Picomolar Inhibitor of Cholesteryl Ester Transfer Protein

Abstract: A novel series of substituted N-[3-(1,1,2,2-tetrafluoroethoxy)benzyl]-N-(3-phenoxyphenyl)-trifluoro-3-amino-2-propanols is described which potently and reversibly inhibit cholesteryl ester transfer protein (CETP). Starting from the initial lead 1, various substituents were introduced into the 3-phenoxyaniline group to optimize the relative activity for inhibition of the CETP-mediated transfer of [3H]-cholesteryl ester from HDL donor particles to LDL acceptor particles either in buffer or in human serum. The be… Show more

“…2003, Reinhard et al (32) described additional members of this series. A 4-chloro-3-ethyl analog was particularly potent, exhibiting an IC 50 in human serum of z50 nM versus 640 nM for SC-795.…”

“…A dataset of 118 N,N-disubstituted-3-amino-2-propanol derivatives (1-118, Table A under Supplementary material) was used for pharmacophore modeling and subsequent QSAR analysis [33][34][35][36]. The conformational space of each inhibitor was sampled utilizing the poling algorithm implemented within CATALYST [21][22][23]37].…”

“…The structures of 118 CETP inhibitors (Table A under Supplementary material) were collected from published literature [33][34][35][36]. The in vitro bioactivities of the collected inhibitors were expressed as the concentration of the test compound that inhibited the activity of CETP by 50% (IC 50 ).…”

“…phenoxyphenyl)trifluoro-3-aminopropan-2-ols of type 33 (Scheme 14), which potently and reversibly inhibit cholesteryl ester transfer protein (CETP) [33]. In most cases, the amino alcohols were converted into amides and eventually oxidized to the trifluoromethylated ketones.…”
