This study aimed to assess the impact of a combination of probiotic strains of Lactiplantibacillus on the treatment of androgenic alopecia (AGA). To this end, 136 individuals with AGA (62 men and 74 women) aged 18–65 years were enrolled in a double-blind, parallel-group clinical trial. A total of 115 individuals (57 in the probiotic group and 58 in the placebo group) completed this study within a 16-week intervention period. Capillary density, thickness, and length of hair were analyzed before and after the intervention using FotoFinder Trichoscale Pro. In addition, the gut microbiota was assessed by paired-end sequencing on the Illumina MiSeq platform (2 × 300 bp). At the conclusion of the treatment period, a notable decline (p < 0.05) in the number of telogen hairs was evident in the probiotic group while hair thickness decreased in the placebo group (p < 0.05). However, the remaining variables did not exhibit any statistically significant changes. In the probiotic-treated group, individuals aged less than 37.5 years exhibited a reduction in the number and density of telogen hair (p = 0.0693 and p = 0.0669, respectively) and an increase in hair length (p = 0.0871). Furthermore, a notable decline in the number and density of vellus hair (p < 0.05) was observed, and this was accompanied by no change in the hair thickness. The probiotic-treated group exhibited a significantly higher abundance of Lactobacillus (p-adjusted < 0.05, DEseq2 test) and demonstrated a notable reduction in the number and density of telogen hair, and this was accompanied by an increase in the percentage of anagen hair. The probiotic mixture was well tolerated by the participants, with a treatment adherence rate of 90%. In light of this study’s limitations, it can be concluded that a mixture of three strains of Lactiplantibacillus promotes the presence of terminal follicles, preventing their gradual miniaturization, which is a characteristic of AGA. The increasing demand for effective hair care products has highlighted the necessity for rigorous claims substantiation methods, particularly for products that target specific hair types. This is essential because the effectiveness of a product can vary significantly based on the hair’s condition and characteristics. A well-defined bleaching protocol is crucial for creating a standardized method to assess product efficacy, especially for products designed to repair damaged hair. The objective of this study was to create a practical bleaching protocol that mimics real-world consumer experiences, ensuring that hair samples exhibit sufficient damage for testing. This approach allows for a reliable assessment of how well various products can repair hair. The protocol serves as a framework for evaluating hair properties and the specific effects of each product on hair structure. Color, brightness, lightness, morphology, and topography were primarily used to understand the big differences in the hair fiber when treated with two repair benchmark products, K18® and Olaplex®, in relation to the Bleached hair. The devised bleaching protocol proved to be a fitting framework for assessing the properties of hair and the unique characteristics of each tested product within the hair fiber. This protocol offers valuable insights and tools for substantiating consumer claims, with morphological and mechanical methods serving as indispensable tools for recognizing and validating claims related to hair. The addition of K18® and Olaplex® demonstrated an increase in hair brightness (Y) and lightness (L* and a*) in relation to the Bleached samples, which were considered relevant characteristics for consumers. Olaplex®’s water-based nature creates a visible inner sheet, effectively filling empty spaces and improving the disulfide linkage network. This enhancement was corroborated by the increased number of disulfide bonds and evident changes in the FTIR profile. In contrast, K18®, owing to the lipophilic nature of its constituents, resulted in the formation of an external layer above the fiber. The composition of each of the products had a discrete impact on the fiber distribution, which was an outcome relevant to the determination of spreadability by consumers. Full article

The monotypic genus Prasiolopsis has been known for a long time, but is often overlooked because of difficulties in identification and the morphological variability between uniseriate filaments and cell packages forming pseudoparenchymatic thalli depending on age. We investigated a strain (SAG 84.81) originally denoted as Trichophilus welckeri, which was isolated from the hairs of the sloth Bradypus tridactylus, and compared it with other available strains of Prasiolopsis and of the sister genus Pseudomarvania. Our investigations clearly showed that this strain differed in morphology, especially of the chloroplast, from those originally described for Trichophilus. Phylogenetic analyses of the SSU and ITS rDNA sequences revealed that the strain SAG 84.81 is sister to several strains of P. ramosa within the Prasiola clade (Trebouxiophyceae). Using the ITS-2/CBC approach, we clearly demonstrated that this strain represented a new species of Prasiolopsis, which we proposed here as P. wulf-kochii. In addition, we evaluated the ITS-2/CBC approach by comparing it with the two species of Pseudomarvania. All investigated strains showed CBCs and HCBCs, which support their species delimitation. The sequencing data of Trichophilus welckeri available in GenBank were phylogenetically re-evaluated by including all representatives of the Ulotrichales (Ulvophyceae). Known for its stimulating effects on the nervous and cardiovascular systems, caffeine has proven remarkable versatile properties. It can be used in a wide range of different products, from anti-aging cosmetics to the pharmaceutical treatment of hair loss. Caffeine is known for its antioxidant properties and is commonly found in moisturising creams recommended as anti-aging or anti-cellulite and also for the treatment of different skin disorders, including androgenic alopecia. This bioactive is also described to be able to enhance the sunscreen scattering effect of well-known ultraviolet (UV) blockers. One of the major challenges remains its penetration capacity into deeper skin layers, which may be achieved by the use of nanosized delivery systems, yet without the risk of transdermal delivery. In this review, we discuss the nutraceutical value of caffeine in cosmetic products, so-called nutricosmetics, which grants this bioactive several advantages in several formulations, in comparison to other potential bioactives of nutricosmetic value. Furthermore, the disclosed effects of bioactives commonly found in coffee, tea, and their by-products are reviewed and discussed. The discussion concludes by highlighting the significant benefits of caffeine in the treatment of skin disorders and its potential to enhance and promote skin health. Full article

Along with their physical health, modern people also need to manage the health of their scalp and hair due to changes in lifestyle habits, job stress, and environmental pollution. In this study, a machine learning model was developed to diagnose scalp conditions such as fine dandruff and perifollicular erythema. Then, transfer learning was conducted using EfficientNet-B0. A web platform that allows users to easily diagnose the condition of their scalp was also proposed. The results showed that the accuracy of the diagnosis model for fine dandruff and perifollicular erythema was 75% and 82%, respectively. It showed good performance in classifying normal, mild, moderate, and severe cases compared to previous studies. Finally, a fast and convenient web platform was developed where users can upload an image and immediately visualize their scalp condition, receive diagnostic results, and see similar cases and solutions. The analysis of user satisfaction indicates that this web application has achieved exceptional outcomes in terms of user satisfaction, garnering high evaluations for its usability, design effectiveness, and overall user experience. This setup enables users to easily check their scalp condition and is accessible to everyone, which is a significant advantage. This is expected to play a crucial role in contributing to global scalp health by advocating the benefits of the early detection and treatment of scalp-related conditions. Full article

Androgenetic alopecia (AGA) is the most common form of hair loss, significantly affecting both men and women worldwide. Characterized by progressive hair thinning and loss, AGA is primarily mediated by dihydrotestosterone (DHT). Recent research has identified numerous single-nucleotide polymorphisms (SNPs) associated with AGA, particularly in genes involved in androgen metabolism, prostaglandin pathways, and vasodilation. These genetic markers offer insights into AGA pathophysiology and potential therapeutic targets. Pharmacogenetics, the study of how genetic variations influence drug response, holds promise for personalized AGA treatment. Identifying SNPs that affect the efficacy of treatments like minoxidil and finasteride enables the development of tailored therapeutic strategies. For instance, genetic variants in the SRD5A2 gene, which affects DHT metabolism, can predict responsiveness to 5-alpha-reductase inhibitors. Beyond pharmacogenetics, RNA interference (RNAi) technologies, e.g., small interfering RNAs (siRNAs), present new therapeutic avenues. Studies have shown the efficacy of RNAi-based treatments in targeting androgen receptors, promoting hair growth in AGA models. Integrating genetic and pharmacogenetic research into clinical practice can transform AGA management, enhancing treatment efficacy and patient outcomes. In conclusion, genetic and pharmacogenetic insights are crucial for developing personalized treatments for AGA, while emerging RNAi technologies offer promising new interventions. These advancements represent significant steps toward more effective and individualized AGA therapies. Full article

Waardenburg syndrome (WS) is a common genetic cause of syndromic hearing loss, accounting for 2–5% of congenital cases. It is characterized by hearing impairment and pigmentation abnormalities in the skin, hair, and eyes. Seven genes are associated with WS: PAX3, MITF, EDNRB, EDN3, SOX10, KITLG, and SNAI2. This study investigates the genetic causes of WS in three familial cases. Whole-exome sequencing (WES) was performed to identify single nucleotide variants (SNVs). Copy number variants (CNVs) were analyzed from the WES raw data and through multiplex ligation-dependent probe amplification (MLPA). The study identified one pathogenic SNV and two novel CNVs, corresponding to type I and type II WS patterns in the three families. The SNV, a nonsense variant (c.1198C>T p.Arg400*), was found in MITF and segregated in the affected father. The two CNVs were a deletion of exon 5 in PAX3 in a family with two affected members and a large novel deletion comprising seven genes, including SOX10, in a family with three affected members. These findings confirmed a WS diagnosis through genetic testing. The study emphasizes the importance of integrating multiple genetic testing approaches for accurate and reliable diagnosis, highlighting their role in improving patient management and providing tailored genetic counseling. Recent studies have highlighted the potential of Mesenchymal Stem Cells (MSCs) as an alternative treatment for Alopecia Areata (AA) due to their immunosuppressive properties. While MSCs have shown promise in cell experiments, their effectiveness in vivo remains uncertain. This study aims to validate local administration of MSC therapy’s efficacy in AA treatment through animal experiments. AA was induced through Interferon-gamma (IFN-γ) administration in mice, and MSC treatment (MSCT)’s effects were assessed visually and through tissue analysis. The MSC-treated group showed more hair regrowth compared to the control (CTL) group. MSCT notably reduced local inflammatory cytokines (JAK1, JAK2, STAT1, STAT3, IFN-γR, IL-1β, IL-16, IL-17α, and IL-18) in AA-induced mice’s skin, but systemic cytokine levels remained unchanged. Furthermore, MSC treatment normalized the expression of Wnt/β-catenin signaling pathway genes (LEF1 and β-catenin) and growth factors (FGF7 and FGF2), which are crucial for hair cycle regulation. This study lays the groundwork for further exploring MSCs as a potential treatment for AA, but more research is needed to fully understand their therapeutic potential. Full article

Carao honey is a potential functional ingredient that can generate added value to food products enriched by reducing waste. In many countries, carao has been utilized for therapeutic purposes since this type of plant extract can inhibit the growth against the most common dermatophytes, a pathogenic fungus that grows on the skin, mucous membranes, hair, nails, feathers, and other body surfaces, causing ringworm and related diseases. The physicochemical characteristics of the cookies were also investigated, which included aW, proximate analysis, hardness, and color. A sensory study was carried out to determine the rejection threshold, acceptability, purchase intent, and general taste of 90 consumers. The gluten-free cookies were prepared with carao honey as a partial substitute for rice flour in different percentages (0%, 2.5%, 5%, and 10%). The carao honey proximate composition, rheological properties, and pasting properties were analyzed. It was emphasized that incorporating carao honey into cookies improves the ingredient’s sustainability. The impact of carao on the physicochemical and sensory properties of cookies was evident in color, flavor, and smell. According to the results, only samples with 2.5% carao were accepted regarding flavor and smell. Overall, liking scores, age, and flavor were the most significant predictors of purchase intent. The information given to panelists did not significantly (p < 0.05) influence liking scores only for cookies. According to physicochemical analysis, carao honey flour was high in carbohydrates (88%). The incorporation of carao produced cookies with darker colors and a softer texture. The study demonstrated that carao flour could be included to produce sensorial accepted cookies at a 2.5% addition level as an alternative in the food industry that benefits from environmental sustainability and, at the nutritional level, improves the availability of nutrients, mainly sugars and proteins. Considering a circular economy perspective, this study evaluates the possible effect of targeted short-term supplementation with Withania somnifera L. (WS; Ashwagandha) on ram’s seminal quality, socio-sexual behaviors, and blood constituents. Black Belly rams (n = 20) received a basal diet comprising feed-leftovers from dairy cows in the north-arid Mexico (i.e., Comarca Lagunera CL). The experimental units, with proven libido and fertility, were homogeneous in terms of age (3.41 ± 0.21 yr.), live weight (LW; 53.8 ± 3.3 kg), body condition (BC; 2.96 ± 0.01 units), initial sperm concentration (2387 ± 804 × 106), and viability (23.9 ± 15.6%). Rams were randomly assigned during the transition reproductive period (i.e., May to Jun; 25° NL) to three treatment groups: non-supplemented control group (CONT; n = 6), low WS-supplemented (LWS; i.e., 100 mg kg LW−1 d−1 × 40 d; n = 7), and high-WS-supplemented (HWS; i.e., 200 mg kg LW−1 d−1 × 40 d; n = 7). The basal leftover diet was offered twice daily (0700 and 1600 h); the experimental period (EP) lasted 47 d. No differences (p > 0.05) among treatments occurred regarding LW and BCS at the onset of the EP. Whereas the greater scrotal circumference (SCRC, cm) arose in the LWS and CONT rams, an increased ejaculated volume (VOLEJA, mL) occurred in the WS-rams. A total of 5/9 (i.e., 55%) appetitive and 3/3 (i.e., 100%) consummatory sexual behaviors favored (p < 0.05) the WS-rams, particularly the HWS rams, towards the final EP. The same was true (p < 0.05) regarding the hemogram variables white blood cell count (×109 cells L−1), hemoglobin concentration (g dL−1), and medium corpuscular volume (fL). This study, based on a rethink–reuse–reduce enquiry approach, enabled connectedness between two noteworthy animal systems in the CL: dairy cows and meat sheep schemes. Certainly, the use of dairy cow feed-leftovers aligned with the short-term supplementation with WS promoted enhanced testicular function, augmented seminal volume, and an increased sexual behavior in Black Belly rams in northern Mexico. Finally, while our research outcomes should enhance not only the resilience and sustainability of sheep production and the well-being of sheep-producers and their families, it may also embrace clinical translational applications. Chemotherapy poses both physical and psychological challenges for patients, prompting many to seek answers independently through online resources. This study investigates German Google search behavior regarding chemotherapy-related terms using Google AdWords data from September 2018 to September 2022 to gain insights into patient concerns and needs. A total of 1461 search terms associated with “chemotherapy” were identified, representing 1,749,312 to 28,958,400 search queries. These terms were categorized into four groups based on frequency and analyzed. Queries related to “adjuvant” and “neoadjuvant” chemotherapy, as well as “immunotherapy”, suggest potential confusion among patients. Breast cancer emerged as the most searched tumor type, with hair loss, its management, and dermatological issues being the most searched side effects. These findings underscore the role of search engines such as Google in facilitating access to healthcare information and provide valuable insights into patient thoughts and needs. Healthcare providers can leverage this information to deliver patient-centric care and optimize treatment outcomes. Full article

Accurate skin diagnosis through end-user applications is important for early detection and cure of severe skin diseases. However, the low quality of dermoscopic images hampers this mission, especially with the presence of hair on these kinds of images. This paper introduces DM–AHR, a novel, self-supervised conditional diffusion model designed specifically for the automatic generation of hairless dermoscopic images to improve the quality of skin diagnosis applications. The current research contributes in three significant ways to the field of dermatologic imaging. First, we develop a customized diffusion model that adeptly differentiates between hair and skin features. Second, we pioneer a novel self-supervised learning strategy that is specifically tailored to optimize performance for hairless imaging. Third, we introduce a new dataset, named DERMAHAIR (DERMatologic Automatic HAIR Removal Dataset), that is designed to advance and benchmark research in this specialized domain. These contributions significantly enhance the clarity of dermoscopic images, improving the accuracy of skin diagnosis procedures. We elaborate on the architecture of DM–AHR and demonstrate its effective performance in removing hair while preserving critical details of skin lesions. Our results show an enhancement in the accuracy of skin lesion analysis when compared to existing techniques. Given its robust performance, DM–AHR holds considerable promise for broader application in medical image enhancement. Full article

The Potential of Resveratrol-Rich Peanut Callus Extract in Promoting Hair Growth and Preventing Hair Loss

Resveratrol has been found to stimulate the growth of human dermal papilla cells (hDPCs) and protect them from oxidative damage. The topical application of resveratrol effectively enhanced hair growth and stimulated the transition of the hair cycle from the resting phase (telogen) into the active growth phase (anagen phase). The aim of this study was to examine the effects of resveratrol-rich peanut callus and sprout extracts on promoting hair growth and anti-hair loss effects in hDPCs and then select the most effective extract (callus) for further assessment in clinical applications. The peanut sprout and callus were produced from plant seeds and extracted with ethanol. Resveratrol was identified as the major constituent in the extracts obtained using the HPLC approach, although no resveratrol was found in the original seed extract. Hair cell proliferation, growth factor expression, and 5α-reductase (5-AR) inhibition assays were used to examine the extracts’ promotion of hair growth and inhibition of anti-hair loss in an in vitro hDPCs model, and standard resveratrol served as a positive reference. Initially, the cytotoxicity of the samples was assessed, revealing that 2.5% of all extracts and 0.02% of resveratrol exhibited a non-cytotoxic effect to hDPCs. The 50% cytotoxic concentration of peanut callus extract (15.50 ± 0.05%) was significantly less cytotoxic to hDPCs than sprout (12.30 ± 0.14%) and seed (11.69 ± 0.26%) extracts (p < 0.05). In vitro experiments indicated that peanut callus extract had the greatest proliferated hDPCs (20.44 ± 0.78%), increased the expression of growth factors (IGF-1, KGF, and HGF) and inhibited 5AR (27.83 ± 2.48%). Then, the callus extract and resveratrol as the positive standard were separately incorporated into a hair serum base and assessed for anti-hair loss effect in a randomized, double-blind, placebo-controlled clinical trial on 60 subjects, and the daily hair shedding count, one-minute combing, and forcible hair pluck tests were monitored for 12 weeks. The clinical study showed that the hair serum containing peanut callus extract and resveratrol were effective in reducing hair shedding (66.98% and 61.47%), decreased hair fall from combing by (83.92% and 68.19%), and increased the ratio of the anagen/telogen by (3.3 and 2.8, respectively). The results suggest that peanut callus extract has a significant amount of resveratrol and the potential to stimulate hair growth and prevent hair loss, making it an alternative bioactive option for cosmetic applications. GhPEL48_Dt, a Pectate lyase (PEL, EC4.2.2.2), is a crucial enzyme involved in cell-wall modification and pectin degradation. Studies have shown that the GhPEL48_Dt also plays a significant role in cotton-fiber development; however, the specific function and regulatory mechanism of GhPEL48_Dt in cotton-fiber development are still not fully understood. Here, we found that the histone deacetylase inhibitor-Trichostatin A significantly reduces the transcript levels of GhPEL48_Dt and its enzyme activity. Further, silencing of GhPEL48_Dt significantly inhibits the initiation and elongation of cotton fibers by promoting pectin degradation, and the heterologous expression of GhPEL48_Dt promotes the development of trichomes and root hairs in Arabidopsis, which suggests that GhPEL48_Dt plays a positive and conserved role in single cell i.e., fiber, root hair, and leaf trichome development. Collectively, this paper provides a comprehensive analysis of the fundamental characteristics and functions of GhPEL48_Dt in fiber development, including the regulatory role of histone acetylation on GhPEL48_Dt, which contributes to the understanding of pectin degradation pathways and establishes a theoretical foundation for elucidating its regulatory mechanism. Androgenetic alopecia is a common disease that occurs in both men and women. Several approved medications have been used to treat this condition, but they are associated with certain side effects. Therefore, use of extracts derived from natural products, such as Siberian sturgeon (Acipenser baerii), and the regulation of the gut microbiota have become important topics of research. Sturgeon is known for its high nutritional value and anti-inflammatory properties; however, its effects on androgenetic alopecia and gut microbiota remain uncharacterized. Here, we aimed to investigate whether solubilized sturgeon oil (SSO) promotes hair growth and regulates the gut microbiome. C57BL/6 mice were divided into four groups. Three groups received topical applications of distilled water, SSO, or minoxidil, and one group was orally administered SSO. Each treatment was administered over 4 weeks. Histopathological analysis revealed a significant increase in follicle number (p < 0.001) and follicle diameter (p < 0.05). Immunohistochemical analysis revealed upregulation of β-catenin and ERK-1, markers involved in hair growth-promoting pathways. Furthermore, microbiome analysis revealed that the reduced gut microbiota was negatively correlated with these markers. Our findings indicate that oral administration of SSO promotes hair growth and regulates the abundance of hair growth-promoting gut microbiota. Full article
