[Role of superoxide dismutase in acute pancreatitis: from antioxidant protection to gene regulation]
Abstract
We present modern data on the role of superoxide dismutase (SOD) in antioxidant protection and gene regulation in acute pancreatitis. Antioxidant enzymes are essential in pathogenesis of numerous diseases. SOD is one of the key enzymes of antioxidant system. In this review, we analyzed activity of this enzyme depending on various factors, mechanisms and role in physiological and pathological processes, in particular, acute pancreatitis. SOD is significantly less active in patients with severe acute pancreatitis accompanied by renal failure, severe circulatory disorders and high mortality. There are some SOD gene polymorphisms, in particular, acute destructive pancreatitis R213G, contributing to acute inflammation. Thus, SOD is not only one of the key antioxidant enzymes, but also potential transcription factor regulating activity of signaling pathways. These aspects can underlie new therapies for diseases.
Представить современные данные о роли супероксиддисмутазы (СОД) в антиоксидантной защите и регуляции активности генов при развитии острого панкреатита. Активность ферментов антиоксидантной системы играет важную роль в патогенезе развития большого количества заболеваний. Одним из ключевых ферментов антиоксидантной системы является СОД. В данном обзоре проанализировано изменение активности данного фермента, индуцированное различными факторами, определены механизмы и роль в физиологических и патологических процессах, в частности при остром панкреатите. Доказано значительное снижение активности СОД у пациентов с тяжелым течением острого панкреатита, сопровождающимся развитием почечной недостаточности, тяжелыми циркуляторными нарушениями и высокой смертностью. Выявлен ряд полиморфизмов гена СОД, в частности острый деструктивный панкреатит R213G, при котором создаются предпосылки для развития острого воспаления. Таким образом, СОД не только является одним из ключевых ферментов антиоксидантной защиты, но и способна выполнять функции фактора транскрипции, регулируя активность сигнальных путей, что может стать патогенетической базой для новых методов лечения заболеваний.
