Objective: This study aims to formulate and characterize the transfersomes for percutaneous delivery of peptides and proteins. In particular, this study was a preliminary study for the transfersomes formulation of recombinant human epidermal growth factor (rhEGF) for topical delivery. Methods: The transfersomes was prepared by thin film hydration method using phosphatidylcholine and sodium deoxycholate as vesicle former. In this study, transfersomes formulas were optimized, namely TF1, TF2, TF3, and TF4 with several ratios of phospholipid and surfactant which were 90:10, 85:15, 80:20, and 75:25, respectively. Afterward, the transfersomes were characterized in terms of particle size distribution, polydispersity index, zeta potential, morphology of vesicles, and deformability index. Results: The results showed that the best formulation was TF3 with the ratio of 80:20 with a particle size of 118.6±1.33 nm, polydispersity index of 0.102±0.011, zeta potential of-30.9±0,46 mV, and deformability index of 1.182±0.08. TEM analysis also showed spherical and unilamellar vesicles of transfersomes. Conclusion: This work demonstrated that the sodium deoxycholate-based transfersomes could be potential to be further formulated with peptide and protein for percutaneous delivery.

Purpose : Recombinant human epidermal growth factor (rhEGF) is a 6045-Da peptide that promotes the cell growth process, and it is also used for cosmetic purposes as an anti-aging compound. However, its penetration into skin is limited by its large molecular size. This study aimed to prepare rhEGF-loaded transfersomal emulgel with enhanced skin penetration compared with that of non-transfersomal rhEGF emulgel. Methods: Three transfersome formulations were prepared with different ratios between the lipid vesicle (phospholipid and surfactant) and rhEGF (200:1, 133:1, and 100:1) using a thin-film hydration-extrusion method. The physicochemical properties of these transfersomes and the percutaneous delivery of the transfersomal emulgel were evaluated. Long-term and accelerated stability studies were also conducted. Results: The 200:1 ratio of lipid to drug was optimal for rhEGF-loaded transfersomes, which had a particle size of 128.1 ± 0.66 nm, polydispersity index of 0.109 ± 0.004, zeta potential of −43.1 ± 1.07 mV, deformability index of 1.254 ± 0.02, and entrapment efficiency of 97.77% ± 0.09%. Transmission electron microscopy revealed that the transfersomes had spherical and unilamellar vesicles. The skin penetration of rhEGF was enhanced by as much as 5.56 fold by transfersomal emulgel compared with that of non-transfersomal emulgel. The stability study illustrated that the rhEGF levels after 3 months were 84.96–105.73 and 54.45%–66.13% at storage conditions of 2°C–8°C and 25°C ± 2°C/RH 60% ± 5%, respectively. Conclusion: The emulgel preparation containing transfersomes enhanced rhEGF penetration into the skin, and skin penetration was improved by increasing the lipid content.

Objective:The objectives of this study were to determine the minimum inhibitory concentration (MIC) of Salam bark extract against Staphylococcus aureus, formulate and evaluate hand sanitizer gels containing Salam bark extract, and determine the effectiveness of the gels against bacteria on the palms of the hands. Methods:The gel base was optimized by preparing three formulations containing carbomer and triethanolamine at ratios of 0.25%:0.5%, 0.5%:1%, and 0.5%:2%. The best gel formulation was mixed with Salam bark extract. The physical stability of gels containing 4.04% (formulation 1) and 7.77% (formulation 2) Salam bark extract was measured at 4±2°C, 27±2°C, and 40±2°C for 12 weeks. The effectiveness of the gels was examined on the palms of 30 respondents. Results:The MIC of Salam bark extract was 3.12%. The best gel base contained carbomer and triethanolamine at a ratio of 1-4 and a pH of 5.50. Formulations 1 and 2 gels had good stability for 12 weeks. Formulation 2 tended to decrease the number of bacteria (p=0.125) better than formulation 1 (p=1.000). In the hedonic study, formulation 2 was preferred to formulation 1. Conclusion:Formulation 2 gel with 7.77% Salam bark extract was more effective than formulation 1 gel with 4.04% extract in decreasing the number of bacteria on the palms.

Background: Andrographolide is a phytoconstituent with anti-inflammatory activity, however, the compound’s poor oral bioavailability has hindered its effective formulation for oral administration. This study, therefore, aims to develop an ethosome for improving andrographolide penetration through the transdermal delivery system. Methods: This study developed 3 ethosome formulas with different andrographolide-phospholipid weight ratios (1:8, 1:9; 1:10), using the thin-layer dispersion-sonication method. Subsequently, the ethosomes were evaluated for particle size, polydispersity index, zeta potential, morphology, as well as entrapment efficiency, and incorporated into a gel dosage form. Subsequently, an in vitro penetration study was performed using Franz diffusion cells for 24 hours and the stability of the gels at 5 ± 2°C, 30 ± 2°C, and 40 ± 2°C, were studied for 3 months. Results: The results showed the optimal formula was E2, a 1:9 weight ratio formula of andrographolide and phospholipid. Based on the transmission electron micrograph, E2 possessed unilamellar, as well as spherical-shaped vesicles, and exhibited superior characteristics for transdermal delivery, with a particle size of 89.95 ± 0.75 nm, polydispersity index of 0.254 ± 0.020, a zeta potential of -39.3 ± 0.82 mV, and entrapment efficiency of 97.89 ± 0.02%. Furthermore, the cumulative andrographolide penetration and transdermal flux for the ethosomal gel of E2 (EG2) were 129.25 ± 4.66 µg/cm2 and 5.16 ± 0.10 µg/cm2/hours, respectively. All the ethosomal gel formulations exhibited improved penetration enhancement of andrographolide, compared to the nonethosomal formulations. Also, the andrographolide levels in the ethosomal and nonethosomal gels after 3 months ranged from 98.13 to 104.19%, 97.93 to 104.01%, and 97.23 to 102.26% at storage temperatures of 5 ± 2°C, 30 ± 2°C/RH 65% ± 5%, and 40 ± 2°C/RH 75% ± 5%, respectively. Conclusions: This study concluded that encapsulation into ethosome enhances andrographolide delivery through the skin.
